The composition of cells is generally divided into four areas: preparation area, preparation area, storage area and sterile area.

1、 Cell culture instrument and introduction


[1. Super clean workbench]


The vertical flow ultra clean bench is generally used for cell culture. Under positive pressure, there is no gas leakage. Only the operating object is protected, but the staff and laboratory environment are not protected.


Ultraviolet irradiation realizes sterilization function. Ultraviolet sterilization (surface) + ozone sterilization (dead corner, baffle closed). Each ultraviolet irradiation lasts for 20-30min, and the air is blown for 10min before use (pay attention to the air change in the culture room). The cleanliness reaches class 100.


After 3000 hours, the power of the ultraviolet lamp will be reduced to 70% of the original. Replace the coarse filter cloth on the upper part in time and remove it regularly for cleaning.


[2. Biosafety cabinet]


Under negative pressure, the gas does not leak out, which not only protects the sample, but also focuses on protecting the operator and the environment. Secondary safety cells carry viruses (such as hele / KB), primary cultured clinical human samples (fungi), lentivirus related experiments.


[3. CO2 cylinder]


CO2 cylinders are mainly used for the storage of CO2 gas. In the process of cell culture, most cells need to be carried out under the condition that the CO2 content is 5%. The CO2 cylinder is mainly connected to the incubator for the stable maintenance of CO2 content.


[4. Inverted microscope]


The inverted microscope is a routine necessary equipment in the daily work of the cell culture laboratory. It is mainly used to understand the growth of cells and observe the occurrence of infection. If funds permit, it is recommended to choose a phase contrast microscope, an anatomy microscope, a fluorescence microscope, a video recording system or a time-lapse film shooting device equipped with a camera system, which can shoot and record the cell growth at any time.


[5. Table centrifuge]


Centrifuge is a special instrument that uses centrifugal force to separate and precipitate mixed liquid (containing solids). It is mainly used to prepare cell suspension, adjust cell density, wash and collect cells. The centrifugation rate in cell culture is generally 300xg (about 1000rpm) for 5-10min. Too high a rotation speed will cause cell death. According to different needs, there are more functional centrifuges to choose from, such as large capacity, adjustable temperature centrifuges, high-speed centrifuges and low-temperature freezing centrifuges.


[6. CO2 incubator]


CO2 is a metabolic product of cells, an essential component of cell growth, and also related to maintaining the pH of culture medium. In cell culture, O2 and CO2 are necessary conditions for cell growth. CO2 incubators have dry and wet types, and the culture conditions are generally set at 37 ℃ and 5% CO2. The water in the water tray of the CO2 incubator is prevented from being contaminated by sterile water + regular 2-week replacement + preservative (0.02% sodium azide or 2% copper sulfate).


[7. Cell counting plate (device)]


The cell counter is a microcomputer controlled counting instrument integrating counting, statistics and display. It is reliable and convenient to use.


[8. Pipette]


The main function of the pipette is to suck and remove liquid such as culture medium during cell culture. The main measuring range used in cell culture consists of 10ul / 100ul / 200ul / 1000ul.
