1. Culture medium

2. Other added ingredients


In addition to the basic nutrients provided by various synthetic media, other components such as serum and factors should be added according to different cells and different culture purposes.


Serum provides important substances such as extracellular matrix, growth factor and transferrin, and fetal bovine serum is commonly used. The proportion of serum added should be determined according to different cells and different research purposes. 10% ~ 20% serum can maintain the rapid growth and proliferation of cells, which is called growth medium; In order to maintain the slow growth or immortality of cells, 2% - 5% serum can be added, which is called maintenance culture medium. However, there are also substances harmful to cell growth and reproduction in serum, such as complement, immunoglobulin and some growth inhibitory factors; The composition is not clear, which affects the analysis of the results; The serum activity of different animals and different batches is different, which affects the stability of the culture effect.


Glutamine is an important nitrogen source for cell growth and plays an important role in the process of cell growth and metabolism. However, glutamine is very unstable in solution and easy to be degraded. It can be decomposed by about 50% after 7 days at 4 ℃, so glutamine needs to be added before use.


In order to prevent contamination, a certain amount of antibiotics should be added to the culture medium.
