The rotating bottle keeps rotating during the culture process, so that the cells can alternately contact the culture solution and air. Rotating bottle culture has the advantages of simple structure, less investment, mature technology, and simply increasing the number of rotating bottles. However, it also has its disadvantages: high labor intensity, large occupation space, small surface area per unit volume for cell growth, low cell growth density, and difficult to control the difference between bottles, so it is difficult to industrialize or scale production.


Microcarrier culture technology is a technology that uses sheet or spherical microcarriers to make cells adhere to its surface and culture in bioreactor. The high-density microcarriers greatly increase the surface area of cell culture, thus realizing the expansion of production capacity. Microcarrier technology also has certain limitations. Only a small number of domesticated cells can be cultured on a large scale in this state. At the same time, due to some characteristics of cell adherent growth, it is difficult to continue to scale up when it is expanded to a certain scale, thus limiting the scope of application of this technology.


Cell factory culture technology is widely used in the vaccine industry, such as hepatitis A vaccine. The technology of cell culture and virus amplification using cell factory has been very mature. In addition, in other vaccine production based on adherent cell culture, such as chickenpox vaccine, mumps vaccine, polio vaccine and other products, the traditional bottle transfer process is gradually replaced. In the process of producing JE and rabies vaccine with bioreactor, the cell factory also plays an irreplaceable role in the rapid and large-scale expansion of cells in its early stage. At the same time, the cell factory also has good applications in cell therapy, monoclonal antibody preparation and other fields. In foreign countries, cell factory culture technology has been a very mature and universal technology for mass culture of cells in vitro. Such as V79-4, chicken embryo fibroblasts, L929, hel299 cell culture, EPO expression, stem cell culture, etc. Due to its convenient and safe operation mode, less occupied space and good controllability, the cell factory has been accepted by more and more domestic production and scientific research users. It has the advantages of being superior to rotating bottles and has no application limitation similar to bioreactor. The application of the cell factory will be more and more extensive, and it will quickly become a relatively mature new generation of cell culture technology in the next few years.
